We are investigating the molecular regulation of genes involved in the acute inflammatory process, interleukin-1b (IL-1b) tumor necrosis factor a (TNFa) and prostaglandin G/H synthase 2 (PGHS 2). Our focus is elucidating transcriptional and translational events responsible for the adaptation of leukocytes to endotoxin-induced expression of the IL-1b and PGHS 2 genes. We seek to identify the cis elements on DNA and RNA that control both the activation of leukocytes and adaptation to endotoxin. Techniques employed in the laboratory include plasmid isolation, DNA sequencing, design constructs, transfecting cells, RNA/DNA isolation, electrophoretic mobility shift and supershift assays, and phosphorylation of signal transduction proteins and transcription factors.
